I have my colors set up completely opposite to yours. My starting point color is white and the others are yellow. But anyway, yes, they will all behave the same. The first vertex is significant though, when it comes time to apply a surface.
The first vertex comes into play when you have several cross-section splines lined up, that you want to skin. The skinning operation is sensitive to the orientation of the first vertex on each cross-section, because the program starts the skin by connecting these first vertices. You want the first vertex of each cross section to be more or less in line with the others. Otherwise, the object may end up looking twisted.
You can change the position of the "First" vertex on a closed Spline by selecting one of the other vertices and clicking the "Make First" button.
