Open your template file and add it to the Drawing Resources folder in the browser. Save and close the template file.

File>New and select the template.

You can also select your template to be the new default when creating a new drawing. The Files tab on the Options menu has a section for Template settings where you can browse out to your template in the "Default Template File Name for QNEW". This way, your drawing template will open by default when you launch AutoCAD or click on the New Drawing button on the toolbar. Not sure if that's what you're after, but it might save you some time if you're using that template exclusively.
